There are two kinds of models to study oxidative stress at home and abroad: animal model and in vitro cell culture model. Among them, the most widely used in vitro cell culture model is H2O2 oxidative damage model. As one of the reactive oxygen species, H2O2 is not only easy to penetrate the cell membrane and react with intracellular iron ions to produce highly active free radicals, but also easy to obtain and stable in nature. It has become an important tool to study all kinds of cell oxidative damage at home and abroad. Technology Description

The establishment of a successful and reliable oxidative stress model is of great significance for screening antioxidant substances and revealing their mechanism of action. Here, we take human hepatoma cell line HepG2 as an example to establish a model. Different concentrations of H2O2 were used to induce different times. By detecting the level of ROS and MDA in cells, as well as the activities of SOD, cat and GSH-Px, the cell model of oxidative stress in vitro was established, which provided a cytological model for screening antioxidant substances and revealing their mechanism of action.

The core step of establishing stress model with hydrogen peroxide is to treat HepG2 cells with different concentrations of H2O2 for different times, and the best scheme of chemical induction has been determined.

The survival rate of cells was detected by MTT-2,5 '- dichloro-diallyl-2, diallyl-diallyl-5 (dichloro-2, diallyl-methyl) blue fluorescence assay, And superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-PX) activities. Finally, it was confirmed that H2O2 induced HepG2 cells to establish the optimal condition of oxidative stress model.
