Oxidative stress is a physiological state in which the concentration of reactive oxygen species (ROS) and antioxidants is unbalanced. Studies have shown that excessive accumulation of reactive oxygen species can lead to cell damage, such as DNA, protein and lipid membrane damage.
Fig. 1 Hydrogen Peroxide and Cell Stress (Ismail et al., 2016)
Peroxide is most active under some oxidative stress conditions, such as H2O2. Hydrogen peroxide is a kind of reactive oxygen species. It is a toxic product produced by normal aerobic metabolism and pathogenic reactive oxygen species. It involves the reaction of oxidase and superoxide dismutase. Hydrogen peroxide is toxic to eukaryotic cells. High doses will lead to the oxidation of DNA, lipids and proteins, leading to mutation and cell death.
To help measure the level of oxidative stress in cells, Creative Bioarray provides specific quantitative analysis services of hydrogen peroxide.
The purpose of our service is to carry out sensitive quantitative detection of hydrogen peroxide or peroxidase. In the presence of horseradish peroxidase (HRP), the probe reacts with H2O2 to produce a colored product that can be detected at 540-570 nm, or a high fluorescence halogenate product read by a fluorescent microplate reader, with an excitation wavelength of 530-560 nm and an emission wavelength of 590 nm (fluorescence format). The peroxidase content in H2O2 or unknown samples is determined by comparing with their respective standard curves. When H2O2 is excessive, the probe can also be used for ultra sensitive determination of peroxidase activity.
We can detect hydrogen peroxide concentrations as low as 0.8 µ m (colorimetry) or 50 nm (fluorescence). This service is applicable to cell lysates, tissue homogenates, cell culture supernatants, plasma, serum, urine or other biological liquids.
In order to achieve better testing results, customers can send samples in any of the following ways.
Sample Type | Requirement | Transportation Conditions |
Tissue | The isolated tissues were made into single cell suspension and added into PBS medium | Normal temperature transportation |
Living cell | Sample size: 1 X 106 cells / test, added into PBS medium | Normal temperature transportation |
Blood | The blood was collected in the anticoagulant blood collection vessel of EDTA heparin sodium | Normal temperature transportation |
