Service Case

Ischemia-reperfusion injury can cause brain injury. Drug X is a candidate drug for the treatment of cerebral ischemia-reperfusion injury. We used H2O2 induced PC12 cell injury to simulate the in vitro model of cerebral ischemia-reperfusion injury, and explored the protective effect and mechanism of drug X.

Experiment design

PC12 cells in logarithmic growth phase were randomly divided into normal group, H2O2 treatment group and candidate drug X low, medium and high concentration groups. H2O2 group and candidate drug treatment group were induced with H2O2 for 24 hours in advance, and then candidate drug groups were administered with different concentrations respectively. H2O2 group and normal plants were cultured in the same volume of medium.

Analyzed parameters

  • PC12 cells were cultured for 24 hours and the cell viability was detected by MTT assay
  • After PC12 cells were cultured for 24 hours, the activities of Caspase-3 and caspase-9 were detected by colorimetry
  • After PC12 cells were cultured for 6 hours, the intracellular ROS content was determined
  • After PC12 cells were cultured for 6 hours, LDH release, MDA content, SOD, cat and GSH-PX activities were detected
  • The expression of bax and bcl-2 protein in PC12 cells was detected by Western blot after cultured for 6 hours
