Oxidative stress refers to a pathological state in which the production rate of reactive oxygen species (ROS) is greater than the clearance rate due to the imbalance of intracellular oxidation and antioxidant system, resulting in cell and tissue damage. Virus infection is closely related to oxidative damage caused by free radical accumulation.

Porcine circovirus type 2 (PCV2) is commonly used in the study of porcine virology and immunology. Macrophages play an important role in both innate and acquired immunity. They are not only important immune cells for the primary defense of the lung but also important target cells for PCV2 infection.

Technology Principles

Some viruses will destroy the balance of the redox state of host cells in the process of infecting and replicating in animal bodies. Therefore, virus infection can cause the accumulation of cellular reactive oxygen free radicals. On the other hand, after virus infection, phagocytes activate and release oxidative cytokines such as TNF-α and IL-1β. PCV2 infected cells can promote the production of ROS and induce oxidative stress, which in turn promotes the replication of PCV2.

We can treat a given macrophage and other cell lines in vivo with porcine circovirus type 2 (PCV2) and establish an oxidative stress model for our customers. Of course, you can also directly purchase the cell model we have established.

For the model that should be customized, we will use the MTT method to detect cell activity, Griess method to detect the level of nitric oxide (NO) secreted by infected cells, DCFH-DA fluorescence probe to detect the level of intracellular ROS, opt fluorescence method to detect the intracellular redox state, reduced glutathione (GSH) content, and spectrophotometry to detect the activities of XOD, MPO and inducible nitric oxide synthase (iNOS).

We will discuss and evaluate the amount and time of PCV2 infection required to establish the model, and provide customers with relevant reports and suggestions accordingly.

Model Application

Infecting immune cells with PCV2 is helpful to explore the relationship between PCV2 infection and cellular oxidative stress, and can provide a reference for the study of its infection mechanism and treatment.
