Creative Bioarray has constructed the oxidative stress model of primary rat hepatic stellate cells (rHSC) and activated human hepatic stellate cell line (LX-2). These two models can basically represent the main oxidative stress molecules in the liver. They will provide effective tools for testing the specific role of drugs in oxidative stress of liver fibrosis and ensure the reliability of the research results. When testing the effectiveness of liver fibrosis drugs, customers can select appropriate cells and oxidative stress models according to different pathological and physiological needs.
The liver is the main place of iron metabolism and the main target organ of iron overload. Iron overload is closely related to the occurrence and development of liver fibrosis. We established an iron overload rHSC oxidative stress model. The oxidative stress model can often be used to test the effectiveness of drugs in the treatment of liver fibrosis in vitro.
We used H2O2 in human hepatic stellate cell line LX-2 to simulate the actual oxidative stress environment in the liver. H2O2 mainly simulates the H2O2 reactive oxygen intermediates widely existing in the liver. Compared with rHSC model, this is a less mild model, and the two models are essentially different in oxidation mechanism.
Hepatic stellate cell activation is the central link of the occurrence and development of hepatic fibrosis. At present, one of the ideas of international research and development of hepatic fibrosis drugs is to find targets from the mechanism of hepatic fibrosis, that is, the activation mechanism of hepatic stellate cells. Then simulating the activation of hepatic stellate cells in different physiological and pathological states in vitro will provide an effective tool for the research and development of liver fibrosis drugs. Oxidative stress is an important factor in the activation of hepatic stellate cells in the occurrence and development of liver fibrosis. Therefore, it is very necessary to construct a scientific hepatic stellate cell oxidative stress model in vitro.
