Skin Fibroblast Oxidative Stress Service

Skin is the largest organ of the body. Skin aging is an important part of aging, which is related to oxidative stress, DNA damage and cell apoptosis. Therefore, preventing and delaying skin aging has become one of the hotspots of life science research. Skin aging includes two processes, intrinsic aging and photoaging.

Fibroblasts are a good model for studying skin intrinsic aging and photoaging.

Creative Bioarray provides our clients with in vitro oxidative stress modeling services for skin fibroblasts. We simulated the photoaging environment by UVA or 8-methoxypsoralen combined with UVA. Hydrogen peroxide induced cell senescence through oxidative stress.

Based on these established models, we can simultaneously provide anti free radical drug / antioxidant administration experiments, pharmacopharmacology, efficacy evaluation and pharmacokinetic analysis. The purpose of this service is to promote the screening and development of antioxidants and anti free radicals in the skin system.

Photoaging Model of Skin Fibroblasts

As an independent disease or syndrome, skin photoaging has become a research hotspot at home and abroad. Simple fibroblasts are a good model for studying skin photoaging.

Modeling service 1

Acute light injury model of fibroblasts induced by long wave ultraviolet (UVA). We sell established stress models and can also provide modeling services for customers.

When the fibroblasts grow to 80%-90% fusion, we will conduct UVA irradiation. After irradiation, we will observe the morphology, cell proliferation activity and SA-β-gal staining was used to test the modeling effect.

Modeling service 2

Photoaging of skin fibroblasts was induced by 8-methoxypsoralen combined with UVA (8-MOP/UVA). We sell established stress models and can also provide modeling services for customers.

When the cultured skin fibroblasts of 2-3 generations grow to 50%-60% fusion, add a certain concentration of 8-MOP for pre incubation for 24h, and then irradiate them with UVA. After the completion of modeling, change the normal culture medium.

After irradiation, we will test the modeling effect by HE staining, MTT, hydroxyproline content, malondialdehyde content and SOD activity analysis.

Intrinsic Aging Model Of Skin Fibroblasts

Aging is a gradual process in which the functions of tissues, organs and cells gradually lose over time. The senescent cell model, namely replicative senescence, can be obtained by continuously culturing human diploid fibroblasts. However, this method takes a long time, and it is difficult to obtain a large number of aging cell populations with the same background in a short time. Therefore, it brings many difficulties to the scientific research related to aging.

As a small molecule oxidant, hydrogen peroxide (H2O2) is easy to cause cell aging by oxidative stress through biofilm system. This is a rapid, convenient and reliable aging cell model.
